Objective To investigate the effect of sodium hydroxybutyrate(GHB-Na) on expression of hippocampal N-methyl-D-aspartate receptor 2A subunit(NR2A) mRNA in hypoxic-ischemic neonatal rat.Methods Postnatal 7 d rat were randomly assigned to control group(NS group),sham group and sodium hydroxybutyrate group(GHB-Na group),GHB-Na group was further randomly divided into GHB-Na 50 mg/kg,100 mg/kg and 200 mg/kg group.Every group was further divided into five subgroups according the time point 2 h,6 h,12 h,24 h,3 d and 7 d after HI insult with n=6 for each subgroup.Under ether anesthesia,the rat pups had left carotid arteries ligated followed by 2 h of hypoxic (8% oxygen) to induce hypoxic-ischemic brain damage.Saline was administered by i.p.injection immediately after HI insult and three times daily thereafter for maximum 7 d for group Sham and group NS.Also,sodium hydroxybutyrate was administered by the same methods in GHB-Na group,and the dosage of sodium hydroxybutyrate was 50 mg/kg,100 mg/kg and 200 mg/kg,respectively.Reverse transcriptase polymerase chain reaction(RT-PCR) was used to analyze the expression of NR2A mRNA semi-quantificationally in the left hippocampus at 2 h,6 h,12 h,24 h,3 d and 7 d after insult.Results At 2 h,6 h,12 h and 1 d after HI insult,the NR2A mRNA in group NS began to gradually decreased and markedly lower than that of group Sham and group GHB-Na(P<0.01).Compared with group Sham,the expression in the group GHBNa was significantly high(P<0.05 or P<0.01)or showed the tendency to increase.At 3 d after HI insult,the expression in group NS increased as compared with that of former time point,but it was still markedly lower compared with Sham group(P<0.05).The expression in group Sham was significantly high compared with GHB-Na50 mg/kg group(P<0.05) or showed increasing tendency as compared with GHB-Na100 mg/kg and GHB-Na 200 mg/kg groups.At 7 d after HI insult,there were no significant difference in the expression among groups(P>0.05).Conclusion Systemically administration of GHB-Na after hypoxia-i

Methods One hundred eighty seven-day(7d) old Sprague Dawley rat pups of either sex weighting 10 g~14 g were randomly assigned to sham group(S), normal saline group(C),GHB 50 mg/kg group(γ50),GHB 100 mg/kg group(γ100)and GHB 200 mg/kg group(γ200),n=6.The procedure for the induction of hypoxia-ischemia was done according to the way described by Rice:under ether anesthesia,pups were subjected to left carotid artery ligated followed by 2 h of hypoxic exposure(O2:N2=8﹪:92﹪).The pups in S group were only aparted left carotid artery which was not obturated and no hypoxia.GHB 50 mg/kg, 100 mg/kg, 200 mg/kg was administered i.p immediately after hypoxia,then a time every 8h inγ50、γ100 and γ200 group respectively.The same volume normal saline was administered i.p in the same way in S and C group.Reverse transcriptase polymerase chain reaction(RT-PCR) was used to analyze the expression of NR1 mRNA semi-quantificationally in the left hippocampus at 2 h、6 h、12 h、1 d、3 d、7 d after insult in S、C、γ50、γ100 and γ200 group. Results The difference of expression of NR1 mRNA between S and C group wasn’t significant.At any time after insult, The differences of expression of NR1 mRNA among C and three GHB group were not significant too.
