Experimental study on a cryosurgery apparatus被引量:3

Cryosurgery is an effective way of curing many diseases including tumors and cancers. It can be applied using a variety of systems and cryogens. Cheap, convenient, reliable equipment still needs to be developed so that cryotherapy may be accepted by surgeons and hospitals. This paper presents a cryosurgery apparatus that utilizes an auto-cascade refrigeration system. Refrigerant mixture R50/R23/R600a was selected as the working fluid. The mixture composition was altered to achieve lower temperatures and higher capacity. The lowest temperature at the cryoprobe could be as low as -100℃, and 8 W refrigeration capacity could be obtained at -80 ℃. An ice ball of 11.6 mm diameter could be formed when the cryoprobe was immersed in a water bath at 37 ℃.

Rapid Communication:Preliminary study on the freeze-drying of human bone marrow-derived mesenchymal stem cells被引量:1

Long-term preservation and easy transportation of human bone marrow-derived mesenchymal stem cells(hBM-MSCs) will facilitate their application in medical treatment and bioengineering.A pilot study on the freeze-drying of hBM-MSCs was carried out.hBM-MSCs were loaded with trehalose.The glass transition temperature of the freeze-drying suspension was measured to provide information for the cooling and primary drying experiment.After freeze-drying,various rehydration processes were tested.The highest recovery rate of hBM-MSCs was(69.33± 13.08) %.Possible methods to improve freeze-drying outcomes are discussed.In conclusion,the present study has laid a foundation for the freeze-drying hBM-MSCs.

二甲基亚砜对羊软骨细胞存活率和SDH活性的影响被引量:1

以羊软骨为实验材料,用不同体积分数的二甲基亚砜(Me2SO)作为保护剂,在4、-20、-30、-70℃温度下研究对羊软骨细胞存活率和琥珀酸脱氢酶(SDH)活性的影响.实验结果表明,Me2SO对细胞的存活率和SDH活性的影响受温度和体积分数的影响较大.在4~20℃时,Me2SO作为单独冷冻保护剂的最佳初始体积分数为10%~20%.在-20^-70℃时,Me2SO作为羊软骨细胞保护剂的最佳应用体积分数为10%~45%.

Optimization study on the rehydration process of lyophilized human platelets被引量:2

Long-term preservation of human platelets will greatly reduce the risk of their shortage. Lyophilization has been proved feasible for this purpose. For the recovery of lyophilized platelets,rehydration is an important process. In this paper,the rehydration proc-esses for 1 mL and 2 mL samples were studied. The effects of prehydration duration(15,30,60,90,120 and 150 min) in 37°C water vapor and the concentration of rehydration solution(25%,50%,75%,100% platelet-poor plasma) on the recovery rate,MPV(mean platelet volume) and PDW(platelet distribution width) were investigated. The mass changes during the prehydration process were weighed. The optimized rehydration conditions are as follows:(1) for 1 mL sample,the prehydration duration was 15 min and for 2 mL sample the prehydration duration was 90 min;(2) the rehydration solution was 75% platelet-poor plasma. Under optimized conditions,the morphology of the rehydrated platelets kept normal and their ultrastructures kept intact,their aggregation capacity to thrombin(1 U/mL) was 82.8% of the fresh ones. These results will be helpful for designing the freeze-drying protocols for human platelets.

Permeation of dimethyl sulfoxide into articular cartilage at subzero temperatures被引量:5

Osteochondral allografting has been proved to be a useful method to treat diseased or damaged areas of joint surfaces. Operational long-term stocks of grafts which supply a buffer between procurement and utilization would contribute to the commercialization or industrialization of this technology. Vitrification has been thought to be a promising method for successful preservation of articular cartilage (AC), but high concentration cryoprotectants (CPAs) are used which may cause high cellular toxicity. An effective way to reduce CPA toxioity is to increase CPA concen- tration gradually while the temperature is lowered. Understanding the mechanism of (31~A permeation at subzero temperatures is important for designing the cryopreservation protocol. In this research, the permeation of dimethyl sulfoxide (MezSO) in ovine AC at subzero temperatures was studied experimentally. Pretreated AC discs were ex- posed in Me2SO solutions for different time (0, 5, 15, 30, 50, 80, and 120 min) at three temperature levels (-10, -20, and -30℃). The Me2SO concentration within the tissue was determined by ultraviolet (UV) spectrophotometry. The diffusion coefficients were estimated to be 0.85×10-6, 0.48×10-8, and 0.27×10-6 cm2/s at -10, -20, and -,30℃, respectively, and the corresponding activation energy was 29.23 kJ/mol. Numerical simulation was performed to com- pare two Me2SO addition protocols, and the results demonstrated that the total loading duration could be effectively reduced with the knowledge of permeation kinetics.

新型低温保存装置性能的实验研究被引量:2

低温保存装置是实现生物材料长期储存的关键设备之一。采用自复叠制冷、自动化控制和最新低温生物技术的冻结线跟踪法开发了一种新型低温保存装置。利用该装置对甘油、丙二醇、二甲亚砜这三种常用的低温保护剂溶液进行了程序控制的变浓度降温和复温实验,使溶液的最终浓度达到玻璃化所需的浓度,最低温度降到-80℃以下。实验表明,新装置性能符合低温保存要求,在生物材料的玻璃化保存领域有良好的应用前景。

二甲亚砜对关节软骨渗透性研究被引量:7

玻璃化被认为是低温保存关节软骨的一种有效方法,但需要考虑低温保护剂渗入软骨的速率以及低温保护剂毒性的制约。本文选用二甲亚砜(DMSO)作为低温保护剂,对其在1℃,22℃和37℃时对绵羊关节软骨的渗透特性进行了研究,得到相应的扩散系数为1.9×10^-6 cm^2/s,3.3×10^-6 cm^2/s和5.7×10^-6cm^2/s,活化能为22.84 kJ/mol。认识低温保护剂对关节软骨的渗透特性有助于建立低温保护剂加入/取出过程的数学模型,从而优化关节软骨的玻璃化保存方案。

噪声应激对大鼠免疫功能的影响被引量:4

以SD大鼠为材料,分别给予80dB和100dB的噪声刺激,在第1、5、10、15、20d检测大鼠的免疫功能的变化.实验结果表明:机体由于噪声应激产生免疫抑制因子,导致T淋巴细胞转化和T淋巴细胞ANAE+率显著下降;巨噬细胞的Fc受体阳性百分率和对抗原的吞噬能力下降;B淋巴细胞的抗体形成及抗体效价降低.表明机体的细胞免疫、体液免疫和非特异性免疫功能下降.
