The synthesis of four analogs of philanthotoxin is described. The preliminary bioassay showed that these compounds all had good insecticidal activities, and the compound 6a had the best killing effect.

In order to increase the cell penetration and decrease the cytotoxicity of the synthetic molecule, we designed and synthesized a DNA-binding ligand, which was a conjugate of polyamide and phospholipid. We examined the differences in the effect on cytotoxicity between the conjugate and the native polyamide. As expected that the in vitro cytotoxicity of the conjugate was obviously lower than that of the native polyamide either in HeLa cells or SH-SY5Y neuroblastoma cells.

A pair of derivatives of tetrameric N-methylpyrrole polyamide were synthesized in order to develop a new method for the study of interaction of the polyamide derivatives with DNA. Indole acetic acid and nicotinic acid were introduced to the polyamide in the synthesized compound, which showed an expected red shift in the UV spectrum. These compounds may function as a potential tool in the detection of the polyamide binding to DNA.
