The endotoxin adsorbents were prepared by coupling divinyl amine ligands on crosslinked chitosan matrix. Their adsorption ability to endotoxin was compared. It was found that the adsorbent with polyethylene polyamine ligand had the highest adsorption capacity and its clearance percent for endotoxin in endotoxemia plasma was over 80%. The results also showed that the adsorbent has good blood compatibility and clinical application foreground.

A series of modified poly(methyl methacrylate) adsorbents were prepared and their adsorption abilities to endotoxin,which is a component of the outer membrance of Gram-negative bacteria,were compared. The results showed that the adsorbent modified by dimethylamine ligand with a long spacer had the best capacity for adsorbing endotoxin. The adsorbent P-N4,having a hydroxyl group at β -site of the ligand,could form the hydrogen bond with phosphate group. The electrostatic interaction,hydrogen bond and cooperative effect of octatomic ring worked simultaneously. All these factors are responsible for the strong interactions between endotoxin molecule and the adsorbent.

Endotoxin is lipopolysaccharide(LPS) derived from the cell membrane of gram-negative bacteria. The concentration of endotoxin in endotoxemia patients′ blood increases quickly and leads to severe sepsis, leading to severe hypertension, cardiovascular continues to have a high failure and death, which is a major cause of death in patients and continues to have a high mortality despite appropriate surgery, potent antibiotic and intensive supportive therapy was conducted. Removal of endotoxin duly and effectually from patients′ blood is very important in clinic. Hemoperfusion can clear toxins from patient′s blood by adsorbent, not only small molecular toxins but also large ones in time. In this paper, the hemoperfusion adsorbent for removing endotoxin was prepared. The physical and chemical parameter of the adsorbent is investigated and the surface structure of adsorbent was investigated through ESEM. The influence of weights of adsorbents and adsorbing time on adsorption ability was also studied. The results showed that the adsorbent with lysine as the ligand had good affinity to endotoxin and the best adsorption capacity of it was 253.1 EU/g when the initial concentration is 10 EU/mL. The blood compatibility of the adsorbent was also studied and the results indicated that the adsorbent has clinic application foreground.

应用计算机模拟的方法研究了内毒素吸附剂的吸附机理.模拟结果显示,以二甲胺为配体的吸附剂,当β位存在羟基时,此羟基可与内毒素分子间形成氢键,并形成一个八元环的稳定结构.此时吸附剂与内毒素之间存在静电、氢键、疏水相互作用和八元环的协同作用.同时模拟了羟基位于配体不同位置的吸附剂与内毒素的相互作用.结果表明,静电作用为主要的相互作用力,羟基的位置对吸附剂的吸附能力影响显著.

以琼脂凝胶为载体,氨基酸为配体,合成了内毒素吸附剂,并对其吸附效果进行了比较.对吸附剂的各种性质进行了表征,选取琼脂-赖氨酸吸附剂对内毒素血症的兔子进行血液灌流,可将血浆中的内毒素降至接近正常值,最高清除率达73.6%,且吸附剂具有良好的血液相容性.

血液灌流技术以其显著的疗效和新颖的治疗方法而发展迅速,目前其应用正日趋广泛,所治疗的疾病已经从药物中毒、肝肾疾病扩展到一系列免疫性疾病。本文综述了血液灌流材料的发展和研究进展,按照种类不同对材料的性质和应用进行了阐述,为血液灌流材料的制备和应用提供参考。
